摘  要:该文制备了AChE/PbO2/TiO2/Ti电流型生物传感器,并用原子力显微镜(AFM)对该电极的表面形貌进行了表征,结合流动注射技术将其用于有机磷农药(OPs)的检测。该文利用有机磷农药对乙酰胆碱酯酶的抑制作用,以硫代乙酰胆碱(ATCh)为底物,实现了对有机磷农药敌百虫的检测。实践表明,PbO2/TiO2修饰的酶生物传感器,具有光电协同催化性能,响应速度快,检测灵敏度高,稳定性好;固定在传感器上的乙酰胆碱酯酶具有良好的酶动力学响应,其表观米氏常数(KM)为1.34mmol/L。对敌百虫的最低检测限达到1.0nmol/L。In this study, PbO2/TiO2/Ti electrode was prepared and investigated with Atomic Force Micrograph (AFM). It was used for the fabrication of acetylcholinesterase (ACHE) immobilized electrochemical sensor combined with flow - injection analysis (FIA) system for the detection of organophosphorus pesticides (OPs), such as trichlorfon. The degree of inhibition of the AChE enzyme by OPs compounds was determined by measuring the electrooxidation current of the thiocholine generated by the AChE catalyzed hydrolysis of acetylthiocholine (ATCh). It was shown that the method of photoelectro - synergistic catalysis offered a fast, sensitive, and stable response. A value of 1.34 mmol/L for the apparent Michaelis - Menten constant ( KM^app) was obtained. The biosensor detected the trichlorfon as low as 1.0 nmol/L.
