摘　　要：目的观察利多卡因对顺铂耳毒性的保护作用。方法将耳廓反射阳性的健康杂色豚鼠30只随机分为3组:(1)对照组:腹腔注射顺铂0.5 mg·kg^(-1)·d^(-1);(2)实验组:腹腔注射顺铂0.5 mg·kg^(-1)·d^(-1),6 h后腹腔注射利多卡因16 mg·kg^(-1)·d^(-1);(3)空白组:腹腔注射生理盐水0.8 mL·kg^(-1)·d^(-1)。连续6 d,第7天检测听性脑干反应(auditory brainstem response,ABR)听闽。耳蜗铺片光镜观察。结果对照组与实验组用药前后同组比较听阈均有所提高,但实验组提高较少,阈值组问比较差异均有统计学意义(P<0.05)。两组毛细胞损伤以第1转为主,分别与空白组相比差异有统计学意义(P<0.01),但实验组较对照组毛细胞缺失率低,组间比较差异有统计学意义(P<0.01)。结论利多卡因对顺铂耳毒性具有保护作用。(中国眼耳鼻喉科杂志,2007,7:350-352)Purpose To investigate the protective effect of lidocaine against cisplatin-induced ototoxicity. Methods Thirty pigmented guinea pigs with positive preyer＇ s reflex were randomly divided into three groups. The animals were administered with intraperitoneal （IP） saline at a dose of 0.8 mL · kg^-1 · d^-l（ blank group）, IP cisplatin at 0.5 mg · kg^-1 · d^-1 alone （control group） and cisplatin in combination with lidocaine at 0.5 mg · kg^-1 · d^-1 6 h later （experimental group） for six consecutive days. The auditory brainstem response（ABR） threshold was recorded before and 6 days after the treatment. At the 7th day, the animals were decapitated and the cochleas were dissected out for surface preparation. Results Animals in both control and experimental group showed an elevation in ABR threshold after the treatment, with a smaller elevation in experimental group（P 〈 0.05 ）. Morphological observations showed a marked loss of hair cells in the first turn of the cochlea in experimental and control group, as compared with the blank group （ P 〈 0.05 ）. However, the experimental group showed a significantly smaller loss of hair cells compared with the control group （P 〈 0.05 ）. Conclusions Lidocaine had a protective effect against cisplatin-induced ototoxicity.
