摘  要:目的探讨大鼠腰段脊髓γ-氨基丁酸(GABA)转运体-1(GAT-1)在神经病理性痛形成中的作用。方法SD大鼠112只,随机分为2组(n=56):假手术组(S组)和慢性坐骨神经结扎损伤组(CCI组)。结扎大鼠左侧坐骨神经建立CCI模型。于CCI前、CCI后1、3、5、7、10、14d随机取8只大鼠,测定机械刺激缩足反应阈值(MWT)和热刺激缩足反应潜伏期(TWL)。于CCI前、CCI后1、3、7、14 d随机取6只大鼠,取腰段脊髓,采用免疫组织化学法检测大鼠脊髓背角GAT-1蛋白的表达。结果与CCI前比较,2组CCI后各时点MWT和TWL均降低(P〈0.05);与S组比较,CCI组CCI后各时点MWT和TWL均降低,CCI后1、3d GAT-1表达升高(P〈0.05或0.01)。结论大鼠神经病理性痛的形成与GAT-1表达增多有关。Objective To investigate the role of spinal cord gamma-aminobutyric acid transporter-1 (GAT-1) in the pathogenesis of chronic neuropathic pain.Methods One hundred and twelve male SD rats weighing 230-270 g were randomly divided into 2 groups (n=56 each):groupⅠsham operation and groupⅡchronic constrictive injury (CCI).In CCI group the rats were anesthetized with intraperitoneal pentobarbital sodium.Left sciatic nerve was exposed and 4 loose ligatures were placed on the sciatic nerve at 1 mm intervals with 3-0 silk suture.In sham operation group the sciatic nerve was exposed but not figated.MWT (mechanical pawwithdrawal threshold) and TWL (thermal paw-withdrawal latency) were measured before (baseline) and on the 1st,3rd,5th,7th,10th and 14th day after CCI.Six animals were killed before and on 1st,3rd,7th and 14th day after CCI in each group and the lumbar segment of the spinal cord was removed for determination of GAT-1 protein expression in the spinal cord by immuno-histochemistrv.Results The MWT and TWL were significantly lower after operation in CCI group than in sham operation group.The GAT-1 protein expression in the spinal cord was significantly increased on the 1st and 3rd day after operation in CCI group as compared with sham operation group.Conclusion The increase in GAT-1 protein expression in the spinal cord plays a role in the development of chronic neuropathic pain.
