葛根素对营养性肥胖大鼠的减肥作用及机制

摘　　要：目的探讨葛根素(puerarin,Pur)的减肥作用。方法建立膳食诱导的营养性肥胖大鼠模型,对肥胖大鼠灌胃给药Pur混悬液6周,观察Pur干预前后体重、肥胖指数、睾肾周围脂肪质量、脂肪系数、脂肪细胞数量、脂肪细胞大小、血脂、血糖及胰岛素(INS)水平变化。结果给予Pur干预后,营养性肥胖大鼠体重、肥胖指数(Lee′s)、睾肾周围脂肪质量、脂肪系数及脂肪细胞大小均明显低于肥胖模型组(P<0.05或P<0.01);高、低剂量组血清甘油三酯(TG)、总胆固醇(TC)含量显著下降(P<0.05或P<0.01)。结论Pur对营养性肥胖大鼠有明显的减肥及改善脂质代谢的作用。OBJECTIVE To investigate the weight-reducing effect of puerarin on obese rats, METHODS After the obese model was built successfully , puerarin was administered intragastrically to obese rats for six weeks . Then , body weight , obese indexes （Lee＇s） ,fat weight around kidneys and testis ,fat index （the ratio of fat weight to body weight） ,adipocyte number ,adipocyte size, blood lipid and serum glucose were detected. RESULTS Body weight ,obese index ,weight of fat tissue ,fat index ,adipocyte size in the high and low dose of puerarin groups were significantly lower than those in obese model group（P 〈0. 05 or P 〈0. 01 ）. Meanwhile, the levels of serum cholesterol and triglyceride were decreased significantly （P 〈 0. 05 or P 〈 0. 01 ）. CONCLUSION Administration of puerarin to rats for six weeks shows weight-reducing effect and improved lipid metabolism.
