生长抑素mRNA在活动期胃溃疡中的表达及意义

摘　　要：目的:检测胃溃疡中生长抑素(somatostatin,SOM)mRNA的表达,探讨生长抑素在胃溃疡发病中的作用。方法:取手术切除的胃溃疡新鲜标本18例,分别在胃溃疡边缘及切端(相对正常胃组织)取材,胃镜胃溃疡标本12例。运用逆转录PCR(RT-PCR)的方法检测两个不同部位中SOMmRNA的表达,其光密度值(OD)与内参照β-Actin的光密度值比较,分析两个部位的OD比值的不同。结果:胃溃疡标本的两个不同部位均有SOMmRNA的表达,在胃溃疡边缘SOMmRNA的OD比值明显低于胃溃疡切端相对正常胃组织的OD值,差异有显著性(P<0.05)。结论:在基因水平证实了SOM在胃溃疡中的表达,而且表明SOM等胃肠激素在胃溃疡的发病过程中起着非常重要的作用。Objective： To investigate the expressions of somatostatin mRNA in the gastric ulcer. To explore the role of them in the pathogenesis of the gastric ulcer.Methods：The 30 gastric ulcer tissues and the 18 normal gastric tissues were studied by using the reverse transcriptase-polymerase chain reaction（RT-PCR）.To compare the differences of the expressions of somatostatin mRNAs.Results：There were the expressions of somatostatin mRNA in the gastric ulcer and the normal tissue. The OD ratio of somatostatin mRNAs were lower in the margins of gastric ulcer than the normal gastric tissues （P〈0.05）. Conclusion： SOM plays important roles in the pathogenesis of the gastric ulcer.
