摘 要:目的应用纳米级羟基磷灰石复合成骨细胞修复兔颅骨缺损,观察新型颅骨修复材料的修复效果,探索临床应用骨组织工程学修复颅骨缺损的可能性。方法体外原代培养兔颅骨成骨细胞,并将其与纳米级羟基磷灰石联合培养。应用联合培养后的复合材料修补颅骨缺损模型,应用大体观察、病理切片及扫描电镜观察修复效果。结果体外培养并鉴定的兔颅骨骨膜成骨细胞可形成钙化结节,ALP染色呈强阳性,具备良好的成骨能力,在新型颅骨修复材料表面生长良好,6周后扫描电镜下观察显示产生大量新生骨组织,达到较满意修复效果。结论新型纳米级羟基磷灰石作为细胞外基质,复合成骨细胞对颅骨缺损进行组织工程学修复是可行的。Objective To repair the cranial defect of rabbit by using the nano-meter cranial defect substitute and osteoconductive materials combining with the cultured osteoblasts in vivo. The effect of the repairing was examined and the possibility of this method for repairing the cranial defect was estimated. Methods The cranial periosteal-derived osteoblasts were cultured by enzyme methods in vitro, then the cells were planted onto the nano-meter cranial defect substitute and osteoconductive materials. Then the combined materials were used to repair the cranial defect model of rabbit, and the results were observed by the morphology, histopathological section and SEM. Results The cultured cells were capable to form calcium node and positive ALP staining, and were successfully planted onto the materials and grow well. The combined materials can form much bone matrix in the cranial defect site. Conclusion As a good extracellular matrix, the nano-meter cranial defect substitute and osteoconductive materials combining with the periosteal-derived osteoblasts can make a tissue engineering bone repair.
