子痫前期患者脐静脉血清对脐静脉内皮细胞人尾加压素Ⅱ(hUⅡ)mRNA表达的影响

摘　　要：目的探讨妊娠高血压疾病患者脐静脉血清对脐静脉内皮细胞人尾加压素Ⅱ(Homo sap iens UrotensinⅡ,hUⅡ)mRNA表达的影响。方法采用人脐静脉内皮细胞株ECV-304,分别加入正常妊娠及子痫前期患者脐静脉血清,培养24h后,用逆转录-聚合酶链反应(reverse transcription-polym erase chain reaction,RT-PCR)测定细胞中hUⅡmRNA的表达。结果子痫前期组的hUⅡmRNA表达(0.636±0.233)高于正常组(0.437±0.188),差异有显著性(P<0.05)。结论妊娠高血压疾病可以引起胎盘组织内皮细胞中hUⅡmRNA表达的增高,提示hUⅡ对该疾病的发生发展可能起重要作用。Objective：To study the effect of preeclampsia umbilical sera on the Homo sapiens Urotensin（hUⅡ） mRNA expression in endothelial cell of umbilical vein.Methods：endothelial cell strain of Homo sapiens umbilical vein（EVC-304） were cultured respectively with umbilical vein sera from normal pregnancies and pregnant women with preeclampsia.After 24 hours,the expression of hUⅡ mRNA was evaluated by reverse transcription-polymerase chain reaction（RT-PCR）.Results：The hUⅡ mRNA expression in preeclamptic group（0.636±0.233）were higher than those in normal pregnancies group（0.437±0.188）,the difference between the two groups was significant（P〈0.05）.Conclusion：The hUⅡ mRNA expression in endothelial cell of placenta could be raised by hypertensive disorder complicating pregnancy,which prompt hUⅡ might play an important role in the development of this disease.
