健脑益智汤对类阿尔茨海默病大鼠血清胆碱酯酶和脑皮质细胞凋亡的影响

摘　　要：目的:观察健脑益智汤对阿尔茨海默病(AD)大鼠血清乙酰胆碱酯酶(AChE)和脑皮质细胞凋亡的影响。方法:老年Wistar大鼠侧脑室注射聚集态的β-淀粉样蛋白25-35(Aβ_(25-35))建立复合式类AD大鼠模型,测定健脑益智汤灌胃给予AD大鼠(10.6,21.2,42.4g·kg^(-1),qd)3周后血清AChE和脑皮质细胞凋亡百分率,实验设立盐酸多奈哌齐(0.525 mg·kg^(-1))作为阳性对照组。结果:与正常对照组比较,AD模型组血清AChE活性增强(P<0.01),脑皮质细胞凋亡百分率显著增加(P<0.01);与模型组比较,健脑益智汤中、高剂量组AChE活性和脑皮质细胞凋亡百分率均降低(P<0.05,P<0.01),与盐酸多奈哌齐组接近。结论:健脑益智汤可抑制AD大鼠血清Ache活性和脑皮质细胞凋亡。Objective：To investigate the effect of Jiannao Yizhi decoction on serum acetyl-cholinesterase （ACHE）activity and cortical cell apoptosis in rats with Alzheimer＇s disease（AD）.Methods：Aggregated Aβ（25-35） was injected into the lateral ventricle of old rats to induce AD.After treatment with ig JiannaoYizhi decoction at 10.6,21.2 and 42.4 g·kg^-1once daily for 3 weeks in AD rats,the serum activity of AChE and apoptosis percent- age of cerebral cortical cells were determined.Results：The serum activity of AChE and cerebral cortex apoptosis percentage was increased in AD rats（P〈0.01）.JiannaoYizhi decoction at 21.2 and 42.4 g·kg^-1reduced the in- creased AChE activity and apoptosis percentage（P〈0.05 and P〈0.01）.The effect of JiannaoYizhi decoction was similar to that of donepezil hydrochloride.Conclusion：JiannaoYizhi decoction can inhibit the serum AChE ac- tivity and cerebral cortical cell apoptosis in AD rats.
