摘　　要：目的研究骨性关节炎（osteoarthritis,OA）软骨组织中白细胞介素-6（interleukin-6,IL-6）的基因表达及其在OA发病机制中的意义。方法选取OA患者6例和创伤性截肢患者5例的膝关节关节软骨组织,HE染色、光镜观察OA软骨组织的病理学特征;提取关节软骨组织的总RNA和蛋白质,RT-PCR和Western印迹杂交方法测定软骨组织中IL-6 mRNA和蛋白质的表达。结果OA软骨组织表面粗糙不平,软骨细胞排列紊乱,数目减少,有细胞成簇现象,可见软骨细胞变性、坏死,软骨厚度变薄,基质染色不均匀,有异染性。本研究中OA患者及健康青年人软骨组织中均有IL-6基因转录和翻译水平的表达;与健康青年组比较,OA患者软骨组织内IL-6 mRNA和蛋白质的表达分别升高88%和160%,两组差异有统计学意义（P〈0.05）。结论IL-6在OA软骨中呈高水平的基因及蛋白表达状态,IL-6基因的差异表达可能是骨关节软骨退变的机制之一。Objective To study the genetic expression of interleukin-6（IL-6）in cartilaginous tissue and nosogenesis in osteoarthritis（OA）.Methods Cartilaginous tissues of knee joint were obtained from 6 patients with OA and 5 patients with traumatic amputation.Characteristic pathobiology of cartilaginous tissue was observed with HE dyeing.The expressions of IL-6 mRNA and protein in cartilaginous tissue were determined by reverse transcriptase-polym chain reaction and Western blot analysis.Results There were characteristic changes in cartilaginous tissue in OA, such as the rough cartilaginous surface,derangement,chondrocyte degeneration and cellular necrosis. The thickness of OA cartilaginous ground-substance was thinning and uneven.The mRNA and protein of IL-6 in OA cartilaginous tissue increased by 88% and 160%,respectively,compared with those in normal cartilaginous tissue（P〈0.05）.Conclusions The gene expression of IL-6 in OA cartilaginous tissue is high.This may play an important part in the cataplasia of cartilaginous tissue in joint.
