摘  要:目的:建立RP—HPLC法测定枸骨叶中羽扇豆醇含量的方法,为枸骨叶的质量分析和评价提供科学依据。方法:采用Agilent Zorbax ODS柱(4.6mm×150mm,5μm),柱温为25℃,乙腈-水(8:2)为流动相,在流速1.0mL·min^-1和紫外检测波长210nm下进样20μL对枸骨叶甲醇提取物进行HPLC分析。结果:羽扇豆醇在0.25~12.7μg(r=0.9998)呈良好的线性关系,平均加样回收率为100.2%(RSD4.3%)。结论:本方法简便,重复性好,为评价枸骨叶药材的质量提供科学依据。Objective: To develop an HPLC method for the quantitative analysis of lupoel in leaves of llex cornuta. Method: The chromatographic separation was accomplished on an Agilent Zorbax ODS column (4.6 mm × 150 mm, 5 μm) eluted with a mobile phase composed of acetonitrile and water ( 8: 2), and with a flow rate of 1.0 mL · min^- 1. The RP-HPLC experiments were achieved at 25 ℃, and UV detection was set at 210 nm. Result: The calibration curve was linear in the range of 0.25-12.7 μg(r =0.999 8) for lupoel, with acceptable precision and accuracy. The method was applied to samples of 1. cornuta from 10 different locations. Conclusion: The established method was proved to be simple, reproducible, accurate, and can be used to evaluate the quality of leaves of 1. cornuta.
