摘 要:目的:研究葛根黄酮在肠道中的吸收部位和吸收机制。方法:通过对大鼠的十二指肠、空肠、回肠和结肠分别进行在体肠吸收实验,用高效液相色谱法测定葛根黄酮主要成分葛根素的含量,计算出葛根素在各部位的表观吸收速率常数Ka和回流4 h的累计吸收药量,从而反映出葛根黄酮的吸收部位和吸收机制。结果:葛根总黄酮中葛根素在十二指肠、空肠、回肠和结肠的表观吸收速率常数分别为0.013 9,0.012 8,0.012 1,0.013 1 h-1;4h的累计吸收药量分别为136.936,122.739,115.230,123.015μg;其在十二指肠段的吸收明显高于其他肠段(P<0.05)。结论:葛根黄酮在大鼠整个肠道中均有吸收;其吸收呈一级动力学过程。Objective: To study the mechanism of intestinal absorption of puerariae flavones from various intestinal segments. Method: The intestine in rats was divided into duodenum, jejunum, ileum and colon. The four parts were cannulated in situ recirculation, respectively. With the HPLC, the contain of puerarin which is the major component of puerariae flavones was measured. The apparent absorption rate constant (K.) and the uptake in 4 hours of puerarin in different segments were calculated and compared. Result: The apparent absorption rate constant of puerarin of puerariae flavones at duodenum, jejunum, ileum and colon were O. 013 9, 0.012 8, 0.012 1,0. 013 0 h^-1, respectively, and the uptake of puerarin in 4 hours were 136. 936, 122. 739, 115. 230, 122. 015 μg, respectively. Compared to the other segments, significant higher percentage of puerarin was absorbed from duodenum after being perfused for 4 hours ( P 〈 0. 05 ). Conclusion : The puerariae flavones can be absorbed in whole intestine and the results indicated that the absorption of puerariae flavones compiled with the first order kinetics.
