老年心律失常病人的普罗帕酮药物动力学

摘　　要：对１１例老年心律失常病人（男６例，女５例，年龄６７±４８岁）进行了普罗帕酮单剂量３００ｍｇ口服药物动力学研究，用ＨＰＬＣ法测定其服药后０．５、１、１．５、４、６、８、１２ｈ血药浓度，经３Ｐ８７软件自动拟合。结果均符合一室模型，平均药动参数Ｌａｇｔｉｍｅ为０．３９±０．１１ｈ，平均消除半衰期为３．７±２．１ｈ，达峰时间为２．９±３．０ｈ，曲线下面积ＡＵＣ为４７８３±４９４３ｎｇ·ｈ／ｍｌ，平均峰浓度为７５８±６７６ｎｇ／ｍｌ。提示老年心律失常病人药物动力学参数与健康志愿者及非老年心律失常病人药物动力学参数基本相似。The pharmacokinetics of propafenone have been evaluated in old aged arrhythmaic patients.11 old aged patients with arrhythmia (M 6,F 5 and age 67±48) were enrolled in the trial.Patients took single dose 300mg of propafenone at the 7:00Am.The propafenone concentrations after 0.5,1,1.5,2,44,6,8,12 hours in serum were measured by HPLC method.The pharmacokinetic parameters were calcuated by one compartment model in all subjects.The mean half life time,lagtime, T max , C max ,and AUC were 3.7±2.1h,0.39±0.11h,2.9±3.0h,758±676 ng/ml and 4783±4943(ng\5h)/ml respectively.The results show that there is nosignificant difference of the pharmacokinetic parameters between old aged arrhythmaic patients,healthy voluteers and non old aged arrhythmaic patients.
