摘 要:研究福辛普利(Fosinopril,FOS)对二肾一狭高血压模型纯种Beagle犬和自发性高血压大鼠(SHR)的降压作用。采用经典的双侧肾动脉狭窄改良手术方法,造成4只肾性高血压犬(RHD)模型,灌胃行慢性交叉性自身对照实验。SHR成年雌鼠80只,完成1次和连续9d灌胃给药2种实验治疗。卡托普利(CAP)为阳性对照药。结果:RHD和SHR中,CAP和各剂量组FOS均出现明显降压作用(P<0.05,或P<0.01),FOS高剂量组降压反应出现早且维持时间长。SHR1次治疗FOS降压作用呈量-效关系,FOS连续给药9d对SHR的治疗呈明显量-效和时-效关系。结论:FOS和CAP均可使RHD和SHR的血压下降,FOS随着剂量的增加其降压作用也增强,相同剂量的FOS对RHD的降压作用优于CAP。CAP和各剂量FOS组对RHD心率无明显影响。To study the effects of fosinopril (FOS) on renal hypertension dog (RHD) and spontaneous hypertension rat.METHODS The FOS therapeutic effcacy was investigated in a two^kidney^one^clipped hypertension model Beagle dog improved in operative method, i.e. RHD, which was arranged in se1fcontrol alternate chronic antihypertension experiments, and in sPOntaneous hypertension rat (SHR) with a single and repeated 9 d administration. Captopril (CAP) was selected as positive control.RESULTS The systolic and diastolic blood pressure (SBP and DBP) of RHD and SBP of SHR in the CAP group and FOS groups were obviously decreased (P< 0.05, or P< 0.01). The decrement of high dose FOS (20 mg. kg-1) group emerged earlier and maintained longer than that of CAP group and low dose FOS (10 mg. kg-1) group. The decreasing SBP effects of a single administration of FoS therapy in SHR groups showed a dose-dependence relationship, and those of repeated administration dividing once a day for 9 d in SHR groups also showed dose-dependence and time-dependence relationship.CONCLUSIONS Fos and CAP can decrease the BP of RHD and SHR. The more dose of FOS, the more effect of reduction of BP. The effect of decreasing BP of FOS is more potential than that of the same dose of CAP- FOS and CAP has no significant effect on rate of RHD and SHR.
