摘 要:目的探讨用无血清培养基进行人头皮体外组织培养的可行性及培养头皮的结构和功能变化。方法用无血清William E培养基进行人头皮气/液界面培养,用解剖显微镜检测毛发生长情况,HE染色法观察头皮组织结构变化,细胞毒性试剂盒检测培养上清液中的乳酸脱氢酶活性。结果头部皮肤经过22天的无血清William E培养基体外器官型培养,仍保持结构的完整性。毛干生长的长度在无血清William E培养基显著高于含血清MEM培养基(P〈0.05)。无血清William E培养基组分别在培养第2天和第16天LDH活性出现两个峰,而含血清MEM培养基在第12天出现峰值。随着培养时间延长,毛囊渐由生长期转变为退行期和休止期。结论这一器官型头皮培养模型可用于头部皮肤及毛囊的研究。Objective To investigate the possibility of culturing human scalp skin in serum-flee medium, and to observe the changes in the function and structure of scalp skin. Methods Intact human scalp skin was obtained from plastic surgery, and cultured at the air/liquid interface in serum-free William's E medium supplemented with penicillin, streptomycin, hydrocortisone, etc. MEM supplemented with serum served as control. Dissection microscopy was used to observe the growth of hairs. The changes of scalp structure were examined by hematoxylin and eosin staining, and lactate dehydrogenase (LDH) activity by cytotoxicity detection kit. Results The human scalp skin maintained an intact structure after a 22-day organ-culture in vitro in William's E medium. The hair growth was more rapid in serun-free William's E medium compared with serum-containing MEM (P 〈 0.05). LDH activity experienced 2 peaks on day 2 and 16, respectively after culturing in serun-free William's E medium, but only 1 peak on day 12 in serum-containing MEM. With the prolongation of culture period, hair follicles gradually turned from anagen to catagen and telogen. Conclusion The organ culture system may serve as a valuable tool for scalp skin and hair research.
