摘  要:目的探讨钛合金颗粒对体外培养的成骨细胞增殖、成骨能力的影响。方法以人骨髓基质细胞(hMSCs)来源的成骨细胞作为研究对象,将不同浓度的钛合金颗粒(1.0、0.1和0.01mg/ml)与其共同培养,分别测量细胞增殖情况,检测碱性磷酸酶(ALP)活性以及I型胶原(COLLⅠ)合成的情况。结果1.0mg/ml组和0.1mg/ml钛合金颗粒抑制了成骨细胞的增殖(P<0.05),0.01mg/ml组对成骨细胞的增殖影响不明显。不同浓度的钛合金颗粒对成骨细胞碱性磷酸酶的活性和I型胶原合成均有抑制作用(P<0.05),存在一定的剂量-时间-效应关系。结论钛合金颗粒对成骨细胞的增殖和成骨能力均有抑制作用,这也是导致假体松动的骨溶解机制之一。Objective To study the effects of particulates titanium alloy on the proliferation, viability of bone-forming of human osteoblasts in vitro. Methods Human bone marrow stormal cells (hMSCs) were obtained and induced to differentiate and proliferate into osteoblasts. The 3rd to 5th generations of human osteoblasts were cultivated with different concentrations of particulates titanium alloy (1.0,0.1 and 0.01mg/ml). Cell growth and proliferation were detected by MTT method. Alkaline phosphatase(ALP) activity and collagen-Ⅰ synthesis were measured by kit. Results The concentrations of 1.0 and 0. 1 mg/ml titanium particle reduced cells proliferation(P〈0.05), whereas 0.01 mg/ml did not statistically change cells proliferation (P 〉 0.05 ). The three different concentrations of particle had effects on ALP activity and collagen-Ⅰ synthesis significantly (P〈0.05) with some degree of dose-and time-effect relationship. Conclusion Titanium alloy particles reduce cell proliferation and suppress cell osteogenesis, which may be one of the mechamisms for the periprosthetic osteolysis.
