血小板膜糖蛋白Ibα基因VNTR多态性与冠心病相关性的研究

摘　　要：目的探讨我国河南汉族人群血小板GPIbα基因巨糖肽区串联重复序列(VNTR)多态性与冠心病的关系。方法选择404例确诊冠心病的患者和500例无心脏病史的健康体检者作对照,应用PCR方法进行VNTR多态性检测。结果扩增产物电泳显示,在中国河南汉族人群中,发现VNTR的6种基因型:AC、BC、BD、CC、CD、DD。本研究未发现VNTR各基因型及等位基因与冠心病的发生间存在明显相关性。P值均大于0.05。与其他基因型相比,在CD基因型中,Gensini冠状动脉评分≥40组与评分<40组间的差异具有显著性,P<0.05,OR=2.3(95%可信区间1.2～4.4)。结论血小板膜GPIbα基因巨糖肽区VNTR的CD基因型与冠心病的严重程度有关。[Objective] To explore the correlation between the polymorphism of platelet membrane GPIbα gene and coronary heart disease in Henan. [Methods] A total number of 404 patients with coronary heart disease （including myocardial infarction and unstable angina） and 500 healthy controls recruited by random sampling with no history of coronary artery disease were investigated by casecontrol study.VNTR polymorphisms in GPIbα were checked by polymerase chain reaction （PCR）. [Result] There were six genetypes of VNTR in Han Nationality of Henan province： AC, BC, BD, CC, CD, DD. It proved no association between any genetypes or alleles and coronary artery province. However, in patients with CD genetype,the differences between Gensini score ≥40 group and Gensini score 〈40 group reached statistics significance, P 〈0.05, odds ratio （OR） is 2.3（95% CI 1.2-4.4）. [Conclusion] It suggests that there is an association between CD genetype of VNTR in platelet membrane GPIbα gene and the severity of coronary artery disease. The results need to be substantiated by a large-scale and prospective study.
