摘 要:通过考察烟叶提取物中游离茄呢醇的含量,为烟叶提取物质量标准的制订提供依据。应用高效液相色谱法,采用DIKma C18色谱柱(150mm×4.6mm,5μm),甲醇-异丙醇(52:48)作为流动相,流速:0.8mL/Mmin,检测波长为210nm,进样量20μL。方法学考察结果表明,茄呢醇的线性范围为0.5-10粥,相关系数r=0.9995,平均加样回收率为98.1%为(n=9),精密度RSD=2.32%;烟叶提取物中游离茄呢醇的含量在8%-12%之间。该方法具有良好的精密度、准确度和灵敏度,以该高效液相色谱法测定烟叶提取物中游离茄呢醇的含量的方法可行。This paper is to determine the content of free solanesol in 19 samples of tobacco abstraction by using HPLC. This method was performed with DIKma C18 column(5μm,150 mm ×4.6mm) ,and the mobile phase was methanol-isopropyl alcohol( V/V = 52:48) ,the flow rate of the mobile phase was 0.8 mL/min,and the UV detection wavelength was 210 nm. Within 0.5-10 μg,the method had a good linerity,r = 0.9995. The average recovery of solanesol was 98.1% (n = 9), with RSD = 2.32%. This method is sensitive and reproducible, and it could be used for quantitative determination of free solanesol in tobacco abstraction.
