二膦酸盐联合IL-2体内诱导卵巢癌外周血γδT细胞增殖的研究

摘　　要：目的探讨二膦酸盐联合白细胞介素-2(IL-2)对体内诱导卵巢癌患者外周血γδT细胞增殖的影响。方法选择山西省肿瘤医院妇科2004年11月至2006年6月收入院的未经任何治疗的上皮性卵巢癌患者28例作为实验组,同期体检健康志愿女性28例为对照组,采用流式细胞仪分别对两组进行外周血总T细胞数、γδT细胞计数测量、比较。实验组行肿瘤细胞减灭术加紫杉醇+卡铂化疗6个疗程常规治疗,4周后测定治疗后外周血总T细胞、γδT细胞计数,并与治疗前进行比较。实验组进行二膦酸盐联合IL-2体外诱导外周血γδT细胞增殖试验,试验阳性患者(20例)行二膦酸盐联合IL-2体内诱导γδT细胞增殖治疗,分别比较诱导前后外周血总T细胞数、γδT细胞计数。结果实验组患者外周血总T细胞数[(1411.6±410.8)/μL]、γδT细胞绝对值[(136.2±114.0)/μL]与相对值[(8.4±5.5)%]均显著低于对照组的(1617.5±488.9)/μL、(175.5±119.1)/μL及(10.2±5.2)%,差异有统计学意义(P<0.05);常规治疗前后卵巢癌患者外周血总T细胞数[(1034.5±336.4)/μL]、γδT细胞计数[(55.7±40.4)/μL]减少,差异有统计学意义(P<0.05);二膦酸盐联合IL-2体内诱导后外周血总T细胞数[(955.6±352.2)/μL、(1415.4±387.4)/μL]、γδT细胞绝对值[(49.2±24.1)/μL、(201.7±53.2)/μL]与相对值[(5.2±1.8)%、(10.1±2.9)%]发生增殖,差异有统计学意义(P<0.05)。结论二膦酸盐联合IL-2可体内诱导卵巢癌患者外周血γδT细胞增殖。Objective To study the effect of bisphosphonat and interlukin2（ IL -2） on inside induction of multiplication of the γδT cell in peripheral blood of the patients with ovary cancer. Methods Twenty - eight untreated patients with ovary cancer who were admitted to Shanxi Tumor Hospital from Nov. 2004 to Jun. 2006 were selected as experimental group. Control group consists of 28 healthy volunteers without obvious gynecologic diseases. Counts of the total T cell and the γδT cell in peripheral blood of experimental group and control group were determined respectively by flow cytometry. For the patients of experimental group, after application of method of tumor extermination, six periods of treatment of taxol ＋ carbo were proceeded and pre - treatment and post - lreatment counts of the total T cell and the γδT cell in peripheral blood were determined after four weeks. For the patients of experimental group, multiplication experiments of the γδT cell in peripheral blood induced by bisphosphonat and interlukin2（ IL- 2） externally were conducted ;for 20 sensitive patients, multiplication experiments of the γδT cell in peripheral blood induced by bisphosphonat and interlukin2（IL-2） internally were conducted,and pre -cure and post - cure counts of the total T cell and the γδT cell in peripheral blood were determined. Result The total T cell,γδT cells absolute value,and γδT cells relative value in ovary cancer were significantly lower than those in the healthy（P 〈0. 05） ; counts of the total T cell and γδT cell in peripheral blood of the patients with ovary cancer changed, but it was not statistically significant （P 〉0. 05）o γδT cells absolute value,and relative value multiplied in pre -cure and post -cure time of bisphosphonat and interlukin2 （ IL - 2）,which was statistically significant （P 〈 0. 05 ）. Cgndusion Bisphosphonat and interlukin2 （ IL - 2） can induce multiplication of the γδT cell in peripheral blood of the patients with ovary cancer internally.
