摘  要:为了筛选较易再生的不结球白菜品种,为转基因工作建立良好的再生体系,以暑绿、苏州青、亮白叶和矮抗5号为材料,研究了基因型、外植体类型以及不同的激素配比对再生频率的影响。结果表明:不同品种中,暑绿的再生频率最高。不同外植体类型中,子叶-子叶柄再生频率最高。种子发芽培养基中添加苯基噻二唑基脲(TDZ)使幼苗生长健壮,利于外植体分化,不定芽分化率均在60.00%以上,子叶-子叶柄在培养基MN+0.5 mg.L-1TDZ+0.5 mg.L-1NAA+7.5 mg.L-1AgNO3中再生频率最高,达到80.00%,表明TDZ在诱导器官的发生中有较强的持续效应。Based on four varieties including Shulü, Suzhouqing, Liangbaiye and Aikang 5 of non-heading Chinese cabbage, effects of genotype, explant and hormone on shoot regeneration were studied to select the easily regenerating variety, and set up better regeneration system for transgenic work. As shown by the result, the regeneration frequency of Shulü was the highest among different cuhivars. The regeneration frequency of petiole with cotyledon was the highest among different explants types. Adding 0. 5 mg·L^-1 thidiazuron (TDZ) into the germination medium could make the seedlings stronger which was beneficial to regeneration, leading to a regeneration rate of over 60. 00% for all mediums. The regeneration rate of petiole with cotyledon was the highest in the medium MN +0. 5 mg · L^-1 TDZ +0. 5 mg · L^-1 NAA +7. 5 mg · L^-1 AgNO3 and the rate reached 80. 00%. The result indicated that TDZ played an obviously persistent role in inducing organogenesis.
