摘 要:目的研究β_2-肾上腺素能受体(ADRB2)基因Gln27Glu多态位点与中国人糖脂代谢及体脂分布之间的关系。方法选取302例上海地区无亲缘关系的中国人,其中糖调节正常者(NGR组)163名,2型糖尿病患者(DM组)139例。采用聚合酶链反应-限制性酶切片段多态性(PCR-RFLP)检测ADRB2基因Gln27Giu多态位点。观察各基因型和等位基因的频率分布及其与糖、脂代谢等临床指标的关系。结果①DM与NGR组的ADRB2基因Gln27Glu变异基因型频率及等位基因频率的差异均无统计学意义(P值均>0.05)。②NGR组中,Glu27等位基因携带者的总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)分别为(5.65±0.218)、(3.97±0.27)mmol/L,显著高于Glu27等位基因非携带者[分别为(5.18±0.19)、(3.30±0.10)mmol/L,P值均<0.05]。结论中国人中存在ADRB2基因Gln27Glu多态位点,其可能与中国人脂代谢相关。Objective To investigate the association of β2-adrenergic receptor gene(ADRB2) Gln27Glu polymorphism with type 2 diabetes and body fat distribution in Chinese population. Methods The ADRB2 Gln27Glu genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) in 302 Chinese subjects in Shanghai, including 163 subjects with normal glucose regulation and 139 with established diagnosis of type 2 diabetes according to the WHO criteria(1999). The phenotypes of the subjects such as the height, body weight, and blood pressure were recorded and the body mass index was calculated. The plasma glucose level, serum insulin and C peptide levels were examined in 75 gram glucose oral glucose tolerance test with also determination of the fasting lipid profiles, and their association with the genotype and allele frequencies was analyzed. Results No significant differences were identified in the genotype and allele frequencies between the diabetic patients and subjects with normal glucose tolerance(both P 〉 0.05). In the latter group, the carriers of Glu27 allele had significantly higher total cholesterol(P 〈 0.05) and higher low-density lipoprotein-cholesterol levels than those who did not carry this allele(P 〈 0.05). Conclusions A possible association is indicated between ADRB2 gene Gln27Glu polymorphism and lipid metabolism in these Chinese subjects.
