摘  要:目的探讨海马源性神经干细胞在分化过程中Eph/ephrin A3通路的表达及其意义。方法神经干细胞提取自新生大鼠的海马区,经分离培养及鉴定,用免疫组化方法检测在神经干细胞分化过程中促红细胞生成素肝细胞受体A3(Eph A3)及其配体ephrin A3的表达情况。结果Eph A3表达于神经干细胞的胞质,随着神经干细胞的分化,Eph A3在胞质中的表达稍有减弱;ephrin A3逐渐在分化细胞的胞质和胞膜中出现并且表达增强。结论在神经干细胞的增殖及分化期存在Eph/ephrin A3通路的表达,二者的跨膜信号传导在神经细胞突起的生长中发挥着一定作用。Objective To investigate the expression and significance of Eph/ephrin A3 signal transduction during the differentiation of hippocampus-derived neural stem cells (NSC). Methods NSCs were cultured from the hippocampus of rat's embryo and identified by immunocytochemistry of nestin. The expressions of Eph A3 and ephrin A3 were detected by immunohistochemistry in the process of the differentiation of NSCs. Results Eph A3 expression was detected in the cytoplasm of NSCs. During the differentiation,the expression of Eph A3 was detected to decrease a little. Meanwhile ephrin A3 appeared gradually in the cytoplasm and the membrane of cells. Conclusion Eph/ ephrin A3 signal transductions are expressed during the proliferation and differentiation of neural stem cells. The membranous signal transduction of them plays an important role in the growth of processes of the cells.
