摘  要:日本金龟子芽孢杆菌是一种在活有机体外很难产生孢子的昆虫病原体。在以可溶性淀粉、酵母膏、磷酸氢二钾、糖类、碳酸钙为主要成分的液态培养基中对影响菌株生长和孢子生成因素进行研究。结果发现,菌体生长的最佳条件是葡萄糖为糖源、培养温度30℃、初始pH值7.6、通气量与菌液体积比1.5∶1、培养时间16h;孢子生成的最佳条件是海藻糖为糖源、温度30℃、pH值8.0、以硫酸锰和大孔吸附树脂为孢子生成促进剂、培养时间24h。在上述条件下,利用两阶段法生产孢子,成功的使孢子在活有机体外的生成率达到70%以上。The spores of Bacillus popilliae which are specialized pathogens of the scarabaeid beetles are difficult to be reproduced in vitro. According to the research on the factors which affect the bacteria's growth and spore-forming in the liquid media composed mainly of starch, glucide, yeast extract, K2HPO4, and CaC03, we found that with glucose as optimum carbonhydrate, at 30 ℃, pH 7.6, ratio of air volume to that of liquid 1.5:1, culture time for 16 h, the bacteria grew best. We also found that the optimal condition for sporulation is at 30 ℃, pH 8.0, culture-time 24 h, and with manganese sulfate and adsorbent resins as sporulation adjuvants, trehalose as carbonhydrate. Under the conditions mentioned above, we adapted the two-stage method, and the result showed that the sporulation rates reached over 70%.
