摘 要:目的探讨中药对拟阿尔茨海默病动物模型小鼠治疗后大脑、肝等组织生化指标的变化。方法用小鼠作为研究对象,以氯化铝拌饲料喂养6.5个月染毒建立模型,以水迷宫时间和错误率表示记忆和判断力的改变。用中药灌胃治疗1.5个月。小鼠分为4个组,即正常组、模型组、治疗Ⅰ组和治疗Ⅱ组。模型组、治疗Ⅰ组和治疗Ⅱ组先用氯化铝染毒5个月,然后两个治疗组分别用乌圆补血口服液和海尔福口服液灌胃,连续45d。实验结束,取大脑、肝组织匀浆和血清测定各项指标。结果实验前后血红蛋白(Hb)各组间差异无统计学意义(P>0.05);脑AChE(乙酰胆碱酯酶)活力(U/mg?蛋白)正常组和治疗Ⅰ组明显高于模型组(P<0.05),正常组明显高于治疗Ⅱ组(P<0.05);脑GSH-Px(谷胱甘肽过氧化物酶)活力(U/mg?蛋白)正常组和模型组明显高于治疗Ⅰ组和治疗Ⅱ组(P<0.01);脑超氧化物歧化酶(SOD)活力(U/mg?蛋白)正常组明显高于治疗Ⅰ组和治疗Ⅱ组(P<0.05或P<0.01);肝GSH-Px(U/mg?蛋白)治疗Ⅰ组明显高于模型组(P<0.05);肝清除率(%)治疗Ⅰ组和治疗Ⅱ组明显高于正常组和模型组(P<0.01);肝SOD活力(U/mg?蛋白)治疗Ⅰ组明显高于模型组(P<0.05);脑丙二醛(MDA)(nmol/mg?蛋白)治疗Ⅰ组和治疗Ⅱ组明显低于模型组(P<0.05);水迷宫时间(s),染毒后3组时间均有延长,其错误率(%),模型组高于其他各组。结论用AlCl3造模组记忆和判断能力明显下降,认为造模成功,而且造模组脑AChE明显低于正常组、治疗Ⅰ组和治疗Ⅱ组,但治疗后脑MDA明显下降,肝、脑清除率明显升高,治疗Ⅰ组肝GSH-Px升高,脑AChE升高接近正常组。治疗Ⅰ组大脑组织出现明显的治疗效果。Objective To study changing of biochemical indices in brain and liver tissue in AD mice after Chinese medicine trealing. Methods As the research object , the mice were fed with AlCl3 for 6 months and a half to build up the model, using the time of water maze and error rate show the change of memory and judgement. Then poured with chinese medicine for one and a half month. The mice were divided into four groups, control group, model group, treated group Ⅰ and treated group Ⅱ. Model group, treated group Ⅰ and treated group Ⅱ were poisoned for 5 month with AlCl3 first, then the two treated groups were poured with Wuyuan oral liquid and haierfu oral liquid respectively, which was lasting for 45 days. After the experiment finished, determined the biochemical indices of brain tissue and liver tissue serum. Results Before and after the experiment ,the content of Hb of every groups have no change ( P 〉 0. 05 ) ; the vitality of AChE in the brain ( U/mg ·prot), control group and treated group Ⅰ were higher than model group obviously ( P 〈 0. 05 ) ;control group was higher than treated group Ⅱ obviously; the GSH -Px vitality in the brain (U/mg· prot) , control group and model group were higher than treated group Ⅰand treated group Ⅱ obviously (P 〈 0. 01 ) SOD vitality in the brain (U/mg · prot), control group was obviously higher than treated group I and treated group Ⅱ (P 〈 0.05 or P 〈 0. 01 ) ; GSH - PX vitality in the liver ( U/mg · prot), treated group Ⅰ was obviously higher than model group ( P 〈 0. 05 ). The clearance of the liver ( % ), treated group Ⅰ and treated group Ⅱ were was obviously higher than model group ( P 〈 0. 01 ) ; SOD vitality in the liver ( U/mg. prot), treated group I was obviously higher than model group ( P 〈 0. 05 ) ; MDA vitality in the brain ( nmol/ mg. prot), treated group Ⅰand treated group Ⅱ were obviously lower than model group (P 〈 0. 05 ) ;The time of the water maze (s) of th
