摘  要:目的探讨核因子-κB(NF-κB)在博莱霉素(BLM)损伤小鼠肺组织培养细胞中的表达,及其对白细胞介素4(IL-4)的影响。方法将15只C57Bl/6小鼠随机均分为3组,对照组和实验组分别经尾静脉注射生理盐水,干预组注射P65反义寡核苷酸。注射处置6h后,实验组和干预组气管内注射博莱霉素-A5(BLM-A5),对照组注射生理盐水。各组小鼠在气管注射后第3天处死,行肺组织原代培养,培养细胞分别进行P65和IL-4免疫组化染色,并进行图像分析,以平均积分光密度(aiOD)表示P65和IL-4的量。两种染色均设立阴性对照。结果对照组、实验组和干预组P65aiOD值分别为:(1.03±0.37)、(462.04±152.25)和(29.38±3.62),P65免疫组化染色的阴性对照值为(0.82±0.41)。实验组P65的表达较对照组及干预组明显增高(均P<0.05),而对照组与阴性对照间差异无显著性(P>0.05)。对照组、实验组和干预组IL-4aiOD值分别为:(29.36±2.00)、(248.19±39.16)和(47.66±5.61),IL-4免疫组化染色的阴性对照值为(0.97±0.37)。实验组IL-4的表达较对照组及干预组明显增高(均P<0.01),对照组与阴性对照间差异有显著性(P<0.01)。实验组小鼠肺组织培养细胞P65的表达与IL-4表达呈正相关(r=0.907;P<0.05)。结论在BLM致肺损伤过程中,NF-κB在肺间质细胞中的表达明显增强,并可能通过间接作用,使间质细胞表达IL-4能力增强。肺间质细胞NF-κB的表达增强可能在BLM致肺损伤病理过程中具有重要意义。[Objective] To investigate the expression of NF-κB and its effect on IL-4 expression in cultured cells from bleomycin(BLM)-injured lung tissue in mice. [Methods] Fifteen C57BL/6 mice (female, 8 wk of age, 17 to 20 g body weight) were randomly divided into 3 groups. The experimental group and the control group were injected normal saline via the tail vein, respectively, but the treatment group was injected antisense oligonucleotids to P65 subunit of NF-κB (900 μg/animal dissolved in 200 μL saline). 6 hours after intravenous injection, the experimental group and the treatment group were treated with BLM-A5 (5 mg/kg dissolved in 20 μL saline) by intratracheal installation, respectively, but the control group was treated with normal saline (20 μL). On day 3 after intratracheal administration, the mice of every group were sacrificed and the cells from lung tissues were cultured. The immunohistochemistry and microscope image analysis were completed to detect the expression of P65 and IL-4 with the average integrated optical density (aiOD). At the same time, the negative controls were set up, respectively. [Results] The aiOD of P65 in the control group, the experimental group and the treatment group were (1.03±0.37), (462.04±152.25) and (29.38±3.62), respectively, the negative control was (0.82±0.41). The expression of P65 in the experimental group increased markedly in comparison with the control group (P 〈0.05) and the treatment group (P 〈 0.05). However, the treatment group was still significantly higher than the control group (P 〈0.01), but there was no difference between control group and negative control (P 〉0.05). The aiOD of IL-4 in the control group, the experimental group and the treatment group were (29.36±2.00), (248.19±39.16) and (47.66±5.61), respectively, the negative control was (0.97±0.37).
