TPZ联合LY294002对人宫颈癌细胞的体外抑制作用

摘　　要：目的研究替拉扎明(TPZ)联合磷脂酰肌醇-3激酶抑制剂(LY294002)对人宫颈癌细胞HELA的体外抑制作用。方法乏氧及空气条件下,TPZ单独及与LY294002联合用药,在不同药物浓度下,作用于HELA细胞株,用MTT法评价其抑癌效果。结果乏氧条件下,TPZ单独及与LY294002联用对Hela的IC50分别为14.2、3.0μmol.L-1,空气条件下为17.5、2.0μmol.L-1。结论TPZ对Hela有抑制作用;LY294002和TPZ联合用药较TPZ独用降低其IC50,LY294002对TPZ有显著增效作用。TPZ对Hela的细胞生长抑制在乏氧与空气条件下无显著差异。OBJECTIVE To study the effect of LY294002, an inhibitor of phosphatidylinositol 3 - kinase, combined with Tirapazamine （TPZ） on the survival of human cervical carcinoma cell line Hela. METHODS Hela cells were treated with TPZ alone and treated with TPZ combined with LY294002 under aerobic and hypoxie conditions, the surviving fraction of the cells was determined by using MTF method.RESULTS The IC50 values of Hela treated by TPZ and TPZ combined with LY294002 were 17. 5 μmol·L^-1 and 2. 0 μmol·L^-1 under aerobic condition, respectively. The IC50 values of Hela treated by TPZ and TPZ combined with LY294002 were 14. 2 μmol·L^-1 and 3- 0 μmol·L^-1 under hypoxie condition, respectively. CONCLUSION TPZ has inhibitory effect on growth of Hela cells. Compared with TPZ treatment, TPZ combined with LY294002 could decrease the IC50 of Hela cells significantly. The differential inhibition effect of TPZ on growth of Hela cells under aerobic and hypoxie conditions was not observed.
