摘　　要：目的诱导人骨髓间充质干细胞向成骨细胞分化。方法体外分离培养人骨髓间充质干细胞,将其分为对照组和诱导组两组。诱导组加入地塞米松、抗坏血酸和β-甘油磷酸,诱导成骨。采用免疫荧光染色的方法检测碱性磷酸酶(AKP)和骨桥蛋白(OPN)的表达;并通过RT-PCR检测骨桥蛋白mRNA的表达;利用硝酸银染色和茜素红染色两种特殊染色方法检测钙盐沉积。结果经诱导,诱导组细胞在呈典型的成骨细胞形态并有钙结节形成;免疫荧光染色结果显示诱导组细胞AKP和OPN均为强阳性表达,RT-PCR结果也显示诱导组细胞骨桥蛋白mRNA表达;两种特殊染色结果显示诱导组细胞有大量的钙盐沉积。而对照组细胞无上述形态改变和表达。结论人骨髓间充质干细胞在一定的诱导条件下能向成骨细胞分化。Objective To study the differentiation of bone mesenchymal stem cells into osteoblast.Methods MSCs were separated from human bone marrow and expanded in vitro. MSCs were divided into two groups： the control group and the induced group. The inductor was composed of Dexamethasone（DEX）, β-phosphoglycerol and Vitamin C. The effects of differentiation into osteoblast were identified with AKP and OPN by immunofluorescence and calcium salt staining.The expression of OPN mRNA were detected by reverse transcription-polymerase chain reaction （RT-PCR）. Results The differentiated cells showed typical osteoblasts, which expressed AKP and OPN, as weU as OPN mRNA. The calcium salt staining showed that there was generous calcium salt in the differentiated cells. Conclusion MSCs could be induced into osteoblasts.
