摘 要:以芦丁(rutin)为对照品,测定了穿心莲提取物中总黄酮的含量。采用荧光光谱(三维荧光、时间驱动模式)和紫外-可见吸收光谱研究了穿心莲提取物与牛血清白蛋白(BSA)间的相互作用,讨论了药物对BSA构象的影响,证实了二者间相互作用为单一的动态猝灭过程,求出了猝灭常数Ksv=0.95×105,并依据能量转移理论确定了药物与蛋白的最近距离r=2.15 nm。The flavonoid in the extract of Andrographis Paniculata (AP) was determined by spectrophotometry with rutin as control sample. The reaction between AP extract and bovine serum albumin (BSA) was studied with 3D fluorescence spectra and ultra-violate spectra, respectively. The effect of AP extract on the conformation of BSA was discussed. The reaction between AP extract and BSA was confirmed to be a single dynamic quenching process. The quenching constant(Ksv = 0. 95×10^5) was acquired and the nearest distance (r = 2. 15 nm) between AP extract and BSA was determined according to the energy transfer theory.
