摘  要:目的观察中药复方"通脉口服液"对体外培养条件下大鼠肾小球系膜细胞(M sC)增殖和分泌IL-1β的影响。方法应用细胞培养技术进行肾小球系膜细胞的传代培养,采用血清药理学方法,制备通脉口服液含药血清、黄芪含药血清、三七含药血清,采用MTT法观察各组含药血清对正常培养条件下及LPS刺激状态下肾小球系膜细胞增殖的影响,ELISA法测定LPS刺激下通脉口服液组细胞上清中IL-1β的含量。结果3组含药血清均可抑制大鼠肾小球系膜细胞的增殖,三组之间比较,通脉口服液组作用要优于黄芪组及三七组。ELISA结果表明:通脉口服液含药血清可以抑制肾小球系膜细胞分泌IL-1β。结论中药复方"通脉口服液"可抑制肾小球系膜细胞的增殖及LPS诱导的系膜细胞IL-1β的产生,肾小球系膜细胞是通脉口服液防治慢性肾炎,延缓肾小球硬化的重要靶细胞之一。Objective To explore the effects of the Tongmai Oral Solution on cell proliferation and Interleukin -1β ( IL -1β ) production in glomerular mesangial ceils(GMC). Methods GMC were cultured with in vitro cell culture technology. Serum containing Tongmai Oral Solution( TM ), Serum containing Huangqi and Serum containing Sanqi were prepared with serum pharmacologi cal methods. Cell proliferation detected by methylthiazolyltetrazolium (MTT) assay and IL -1β level by enzyme - linked immunosorbent assay(ELISA) were used to evaluate the effect of TM -containing serum , Huangqi -containing serum and Sanqi - containing serum. Results TM - containing serum, Huangqi - containing serum and Sanqi - containing serum could inhibit the proliferation of mesangial cell, and the inhibitory effect of TM - containing serum was stronger. Protein of IL -1β was increased significantly in the presence of LPS and this increase could be reversed by TM - containing serum. Conclusion The glomerular mesangial cell is one of the main target cells which the Tongmai Oral Solution brings therapeutical action and the mechanisms may be related to inhibiting the mesangial cell proliferation , decreasing the IL -1β secretion, and blocking the inflammatory action in mesangial region.
