摘 要:以7个微卫星标记,PCR扩增,12%非变性聚丙烯酰胺凝胶电泳,sanguinetti银染法显色,对四川建昌、金堂、乐至、合江、江安、自贡、嘉陵、营山、白玉9个黑山羊品种(群体),进行X染色体微卫星DNA遗传多态性研究。结果表明:7个微卫星座位在9个黑山羊品种(群体)中均为高度多态座位,建昌、金堂、乐至、合江、江安、自贡、嘉陵、营山和白玉9个黑山羊的平均PIC、H和Ne分别为:0.7644,0.7966,5.1145;0.7258,0.7628,4.4660;0.7438,0.7769,4.7242;0.7580,0.7910,5.0066;0.7365,0.7747,4.5084;0.7463,0.7819,4.6944;0.7683,0.7993,5.1948;0.7642,0.7956,5.0333;0.7211,0.7600,4.2867。嘉陵与营山黑山羊在D=0.284处聚为一类;自贡与江安黑山羊在D=0.294处聚为一类;金堂与乐至在D=0.381处聚为一类,在D=0.395处再与合江黑山羊聚为一类;白玉与建昌黑山羊在D=0.456处聚为一类。最后4种聚为一大类。Genetic diversity for Sichuan black goat populations, including Jintang black goat , Lezhi black goat, Jianchang black goat, Hejiang black goat, Jiangan black goat, Zigong black goat, Jialing black goat, Yingshan black goat and Baiyu black goat, were estimated using seven microsatellite makers by PCR, polyacrylamide gel electrophoresis and silver staining of Sanguintti method. The results showed that seven microsatellite loci analyzed were highly polymorphic loci in nine populations.
