影响短链DNA电泳结果的有关因素分析

摘  要:在电泳制备N-ras第一外显子DNA(0.8kb)的实验中发现:在环境温度为6℃左右时,2kb以下的DNA电泳带出现明显的弥散及拖尾现象。在排除诸多因素后我们最终证实:较短的线性DNA在进行琼脂糖电泳时,除了受文献中所述的样品加样浓度、电泳缓冲液的类型、电泳电压的高低以及溴乙锭的嵌入等因素影响外,电泳时的温度也是影响电泳结果的一条重要因素。将电泳温度提高至20℃时,电泳带弥散及拖尾的现象即可消除。A problem had been encountered when we were preparing the first exon DNA of N ras (a fragment about 0.8 kb) by theprocess of agarose gel electrophoresis that remarkably smearing and tailing bands were observed with the DNA fragments shorter than 2 kb when the experiment was done at 6℃. With the exclusion of all other possible factors, we found out that temperature was a very important factor affecting the migration of small linear DNA molecules, besides the concentration of the sample DNA, the composition of the electrophoresis buffer, the voltage applied, the presence of intercalating dyes and so on. The smearing and tailing phenomena disappeared when the gel was running at 20℃.
