摘  要:[目的]研究氯丹对大鼠甲状腺激素水平及组织结构影响的时间效应。[方法]21d龄SD大鼠检疫1周后,以15mg/kg的剂量经口给予氯丹,分别染毒5、10、15、20、30、40d,对照组给予相同体积的玉米油处理相同的时间。用放射免疫方法测血清甲状腺激素含量,HE染色观察甲状腺组织结构改变。[结果]血清FT4、FT3和TSH的浓度d5时未发现变化;d10、15时FT4、FT3水平下降,伴有TSH水平升高;d20、30、40时FT4、FT3和TSH的浓度又恢复正常。组织病理学观察发现,d10 ̄15时甲状腺滤泡上皮增生成复层;d20甲状腺泡上皮继续增生,有的滤泡局灶性发展到3 ̄4层的增生斑;d30、40时已可见实心芽和实心继发滤泡。[结论]在一定剂量条件下,氯丹可干扰机体内甲状腺素水平,改变其组织结构,并存在一定的时间-效应关系。[Objective] To study the time-dependant effects of chlordane on thyroid hormones and histological structure in rats. [Methods] 120 healthy 21-day-old SD rats were orally given chlordane at the dose of 30mg,/kg after quarantining for a week, and were used narcotics by 5, 10, 15, 20, 30 and 40 days, respectively, and the control group were supplied with equal volume of corn oil with the same time. The contents of thyroid hormones were measured with radio-immunity method, while the histological structure of thyroid gland were observed by dyeing with hematoxylin-eosin (HE). [ Results] The concentration of FT4, FT3 and TSH in serum at 5 days did not found any change, and the concentration of FT4, FT3 were observed to decline at 10 days and 15 days, while the concentration of TSH showed to increase. The concentration of FT4, FT3 and TSH were observed to become normal at 5 days, 10 days and 15 days. By histopathology, thyroid follicle epithelium of chlordane proliferated into multiple-layer structure at 10 days and 15 days; at 20 days, the thyroid follicle epithelium of chlordane continued to proliferating, and some follicle focal developed to proliferative plaque with 3 to 4 layers. Solid bud and follicles secondary, to the solid bud could be detected at 30 days and 40 days. [Conclusion ] Chlordane could affect level of thyroid hormones at a certain dosage and change its histological structure with time-effect relationship.
