摘 要:目的观察大鼠皮质神经元细胞在缺氧/缺糖(OGD)时线粒体膜电位(MMP)的变化及阿司匹林的保护作用。方法取体外培养7d的Wistar大鼠皮质神经元细胞,随机分为正常对照组、缺氧/缺糖模型组、缺氧/缺糖加阿司匹林组。缺氧/缺糖2h后在常氧下继续培养24h。流式细胞术检测不同时间段神经元细胞线粒体膜电位。结果缺氧/缺糖损伤2h,缺氧/缺糖组线粒体膜电位水平较正常对照组显著降低(P<0.01)。缺氧/缺糖加阿司匹林组皮质神经元细胞在缺氧2h及再复氧24h后细胞线粒体膜电位明显高于缺氧/缺糖模型组(P<0.01)。结论阿司匹林可抑制缺氧/缺糖损伤所致的线粒体膜电位的降低,从而具有稳定线粒体膜电位的作用,抑制神经细胞凋亡的发生。Objective To investigate the effect of oxygen/glucose deprivation (OGD) on mitochondrial membrane potential in cortical neurons of rat and the protective effects of aspirin. Methods Cortical neurons cultured for 7d were randomly divided into three groups: Control group, OGD group and OGD with aspirin treatment groups. The mitochondrial membrane potential of neurons was measured with flow cytometry. Results OGD caused significant decrease in the mitochondrial membrane potential in cortical neurons. Aspirin could increase the mitochondrial membrane potential. There was significant difference between the OGD and OGD with aspirin groups (P〈 0.01 ). Conclusion Aspirin could ameliorate the damage of neurons caused by OGD. Aspirin had definite protective effect on cortical neurons.
