摘 要:目的:研究黄芪多糖对大鼠局灶性脑缺血再灌注损伤的治疗保护作用。方法:利用线栓法阻断大脑中动脉(MCAO)制作大鼠局灶性缺血再灌注脑损伤模型,并将实验大鼠随机分假手术组,模型组,黄芪多糖低、中、高剂量组,于再灌后第1天、第3天、第7天处死取材,检测各组大鼠脑组织含水量、超氧化物歧化酶(SOD)、丙二醛(MDA)变化情况,辅以氯化三苯四氮唑(TTC)染色观察脑梗死灶变化情况。结果:与假手术组比较。模型组脑组织含水量及MDA含量显著升高,梗死灶体积显著增大,SOD值明显降低;与模型组比较,黄芪多糖各剂量组的脑含水量,MDA含量显著降低,SOD值显著增高,梗死范围也明显减小。结论:黄芪多糖可以通过降低脑组织含水量、MDA水平,升高SOD水平,对缺血脑损伤有一定的保护作用。Objective:To study the protective effect of Astragalus polysaccharide on focal cerebral ischemia-reperfusion in jury in rats. Methods:The models of focal brain ischemia-reperfusion injury was prepared by middle cerebral artery occlusion(MCAO) in SD-rats. The rats were randomly divided into sham operation control group, model group, low, middle and high dose drug group, after treatment the brain water content and the level of SOD, MDA were measured at the first, third and seventh day after reperfusion. The range of cerebral inflarct was observed by TTC dyeing. Result:Compared with the sham operation control group, the brain water content and the level of MDA were significantly higher, and the level of SOD was significantly lower in model group. The brain water content and the level of MDA were significantly lower, and the level of SOD was obviously higher in low, middle, high dose groups than ones in model group. Conclusion:Protective effect of Astragalus polysaccharide on focal cerebral ischemia-reperfusion in jury in rats was remardable by reducing the brain water content and the level of MDA, and increasing the level of SOD.
