摘 要:目的建立桑叶中总生物碱和1-脱氧野尻霉素(DNJ)含量的测定方法,并考察不同采摘时间、不同种植基地及药用部位的桑叶药材中总生物碱和DNJ含量。方法采用雷氏盐比色法测定总生物碱含量;应用HPLC-ELSD测定DNJ含量。流动相为乙腈-水(80:20);流速:1.0mL.min-1;柱温30℃;进样量:20μL。漂移管温度55℃;载气(N2)压力:0.15MPa。结果本实验所建立的方法准确、重复性好。不同采摘时间、采摘部位的桑叶中二者含量差异较大,不同种植基地的桑叶中二者含量变化相对较小。结论测定结果表明,桑叶中DNJ和总生物碱的含量与采摘时间、种植基地及药用部位等密切相关,其中采摘季节、部位影响较大。OBJECTIVE To establish a method for the determination of the total alkaloids and 1-deoxynojirimycin (DNJ) in Folium Mori and compare the content of alkaloids and DNJ in Folium Mori with different collection time,species,plant site and medicinal part. METHODS The content of alkaloids was measured by Spectrophotometry based on the reaction of alkaloid with Remecke salt and the contents of DNJ was determined by HPLC-ELSD. The HPLC analysis was performed on Agilent ZORBAX NH2 column (4. 6 mm ×250 mm,5 μm ), the mobile phase was MeCN-H2O ( 80:20 ), the flow rate was 1.0 mL ·min^-1, the temperature of column was 30 ℃ and the injection volumn was 20 μL. The ELSD condition were as follows: the temperature of drift tube was 55 ℃, and the gas pressure was 0. 15 MPa, and the value of gain was 7.0 . RESULTS The methods developed were accurate and repeatable. The contents of total alkaloids and DNJ varied obviously among Folium Mori from different collection time and medicinal part,while no significant difference was found in Folium Moil from different plant sites. CONCLUSION The contents of total alkaloids and DNJ correlated with collection season, plant site and medicinal part, and collection season, species and medicinal part were the major factors.
