摘  要:目的:研究鸡矢藤提取物(EPS)对尿酸钠晶体诱导大鼠急性痛风性关节炎(GA)的影响。方法:采用尿酸钠晶体制备大鼠GA模型,用足爪容积测量仪测定造模后不同时间大鼠的踝关节肿胀度;观察造模24h后大鼠步态并进行行为学评分;以HE染色观察大鼠关节组织的病理学改变;采用放射免疫分析法测定大鼠关节组织中白介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)的水平。结果:EPS能显著降低GA模型大鼠的踝关节肿胀度,改善其步态异常。病理组织学检查结果显示,EPS可减轻GA模型大鼠踝关节的组织水肿,减少关节组织炎性细胞浸润,以及改善滑膜增生等病理改变。放射免疫分析检测结果显示,EPS可显著降低GA大鼠滑膜组织中TNF-α和IL-1β的水平。结论:EPS对尿酸钠晶体诱导大鼠GA有显著的改善作用,其机制可能与抑制TNF-α和IL-1β水平有关。OBJECTIVE: To study the effect of the extracts of Paederia scandens (EPS) on acute gouty arthritis induced by monosodium urate in rats. METHODS: Monosodium urate crystals were used to establish gouty arthritis model in rats, with ankle joints' swelling degree at different time after modeling measured using paw volume measuring meter and gaits of rats scored at 24h after modeling. Pathological changes of articular tissues of rats were observed with HE staining and TNF - α and IL- 1β levels were monitored by Radioimmunoassay. RESULTS: EPS could significantly inhibit joint swelling of rats and improve the gaits; histopathologically, EPS could abate tissue swelling of ankle joints, reduce inflammatory cell infiltration of articlar tissues and improve the pathological changes of synovial membranes. Radiommunoassay revealed that EPS could reduce levels of TNF - α and IL - 1β in rats' synovial membranes. CONCLUSION: EPS have remarkable efficacy in improving symptoms of acute gouty arthritis induced by monosodium urate crystals in rats, which might be related to its inhibition on levels of IL- 1β and TNF - α.
