摘 要:目的探讨不同分化程度口腔鳞癌细胞FAS蛋白的表达及其同鳞癌细胞凋亡的关系。方法采用免疫组织化学方法检测38例不同分化程度口腔鳞癌组织FAS蛋白的表达,脱氧核糖核酸末端转移酶介导的原位末端标记(TUNEL)技术,检测鳞癌细胞的凋亡。结果FAS蛋白表达及癌细胞的凋亡与口腔鳞癌的分化程度有关,低分化鳞癌的FAS蛋白阳性表达率显著低于高、中分化鳞癌(P<0.05),癌组织分化程度越低,癌细胞凋亡指数(AI)也越低(P<0.01);口腔鳞癌细胞FAS蛋白表达阴性组癌细胞AI明显低于FAS蛋白表达阳性组(P<0.01),FAS蛋白表达阳性组中,随FAS蛋白表达的下降,癌细胞AI逐步降低,凋亡指数与FAS蛋白的阳性表达呈正相关,各组间AI差异有统计学意义(r=0.68,P<0.01)。结论口腔鳞癌细胞FAS蛋白表达与癌组织分化程度及癌细胞凋亡有密切关系,FAS蛋白诱导的凋亡异常在口腔鳞癌的发生发展中起重要作用,FAS蛋白检测可能作为临床判断口腔鳞癌恶性度的有用标记物。Objective To study the relationship between FAS expression and apoptosis in different differention level Oral Squamous Cell Carcinoma (OSCC). Methods The expression of FAS protein of OSCC and apoptosis of tumor cells were detected by immunohistochemical method and terminal deoxynucleotidyl transferase labeling technique (TUNEL) in 38 cases of OSCC. Results The positive rates of FAS in well and middle differentiated OSCC were higher than those in poor differentiated OSCC (P 〈 0.05 ). The apoptotic index (AI) of tumor cells in FAS positive OSCC was remarkably higher than that in FAS negative OSCC (P 〈0.01 ). There was positive correlation between expression FAS protein and cell differentiation and apoptosis in OSCC ( r = 0. 68, P 〈 0.01 ). Conclusion The expression of FAS was highly correlated with the differentiation and apoptosis in OSCC. It is important for FAS antileptic apoptosis abnormality in OSCC. FAS protein detection is a possible useful tumor marker for clinical judgment of differention level of OSCC.
