摘 要:目的探讨再生障碍性贫血(简称再障)小鼠外周血单个核细胞Th1细胞转录因子T-bet的表达及骨髓巨噬细胞对正常小鼠外周血单个核细胞T-bet的影响。方法①免疫介导(γ射线照射和淋巴细胞输入)的方法建立再障小鼠模型。②RT-PCR方法检测再障小鼠外周血单个核细胞上T-bet mRNA的表达。③正常小鼠外周血单个核细胞分别经再障小鼠骨髓基质细胞培养上清和小鼠骨髓型正常巨噬细胞株Ana-1培养上清刺激后,用RT-PCR方法检测T-bet mRNA的表达。结果①再障小鼠外周血单个核细胞T-bet mRNA的表达(1.49±0.18)较正常对照组(1.03±0.06)增高(P<0.05);②正常小鼠外周血单个核细胞分别经再障小鼠骨髓基质细胞培养上清和Ana-1细胞株培养上清刺激后,T-bet mRNA的表达分别为1.29±0.03和1.54±0.03,均较正常对照组(1.03±0.06)增高(均P<0.05)。结论结果提示巨噬细胞可能通过增加T-bet的表达参与再生障碍性贫血的免疫发病机制。Objective To study the expression of transcription factor T-bet of T helper 1 type cells in peripheral blood mononuclear cells(PBMNC) in aplastic anemia mice, and to explore the effect of mice bone marrow macrophage on T-bet in PBMNC of normal mice. Methods (1)Immunologically mediated method was used to establish the model of aplastic anemia in BALB/C mice. (2)The expression of T-bet mRNA in PBMNC in aplastic mice was measured by reverse transcriptase polymerase chain reaction(RT-PCR). (3)PBMNC in normal mice was stimulated by supernatant of cultured myeloid matrix cells in aplastic mice and mouse bone marrow macrophage cell lines Ana-1 respectively, and the expression of T-bet mRNA was measured by RT-PCR. Results (4)The expression of T-bet mRNA in PBMNC in aplastic mice ( 1.49 ± 0.18) was higher than in normal mice ( 1.03 ± 0.06 ) ( P 〈 0.05 ). (2)Stimulated by supematant of cultured myeloid matrix cells in aplastic mice and mouse bone marrow macrophage cell lines Ana-1, the expression of T-bet mRNA in PBMNC of normal mice was 1.29 ± 0.03 and 1.54 ± 0.03, respectively, higher than that of normal controls ( 1.03 ± 0.06) ( P 〈 0.05). Conclusion The results indicate macrophage may participate in immune pathogenesis of aplastie anemia through increasing the expression of T-bet.
