摘  要:目的探讨山苦茶提取物(Mallotus furetianus)对动脉粥样硬化(Atherosclerosis,AS)大鼠血管内皮功能的影响。方法梯度浓度稀释山苦茶B、D,与DDPH反应,测自由基清除率及其EC50。一次性给大鼠腹腔注射VD370万U/kg,高脂饮食,建立AS模型,并预防性给予山苦茶B、D处理。9周后处死大鼠,取血清测定SOD、NOS、MDA。迅速取出胸主动脉,制成宽3mm的主动脉环,保留内膜,并测定其内皮依赖性舒张能力。结果山苦茶可以清除自由基(山苦茶D>维生素C>山苦茶B),并可明显降低AS大鼠血清MDA水平,提高NOS、SOD活性,增强内皮依赖性血管舒张能力。结论山苦茶B、D增加AS大鼠内皮依赖性血管舒张能力,可能与增强NOS活性、清除自由基、提高机体抗氧化能力有关。Objective To investigate the effects and mechanism of extract of Mallotus furetianus on endothelial function of artery in atherosclerotic rats. Methods The ability in clearance of 1, 1 - diphenyl - 2 - picrylhydrazyl free radical (DDPH). VD3 by Mallotusfuretianus was observed and high - fat diet were given to rats to make atherosclerotic model, different parts of Mallotus furctianus were administered to prevent the development of atherosclerosis. Mter 9 weeks, rats were killed and the blood sample was collected to determine the level of MDA and the activities of SOD, NOS. Vascular circles were made to measure endothelium dependent vascular relaxation. Results The extract of Mallotus furetianus was capable of removing DDPH free radical, and reducing MDA, enhancing the activities of SOD, NOS, improving the ability of endothelium dependent vascular relaxation. Conclusion The extract of Mallotus furetianus can improve the ability of endothelium dependent vascular relaxation in atherosclerosis rats, this effect is possibly associated with the ability of clearance of free radical, increasing activity of NOS, and the enhanced antioxygen capability.
