摘  要:目的观察更年安怡片对绝经后骨质疏松雌鼠预防和治疗的效应及其作用机制。方法雌性SD大鼠60只,随机分为正常组(INT),假手术组(Sham),模型组(Model),雌激素组(E2),更年安怡片组(GNAT)和更年安怡片预防组(GNAT PR),每组10只。检测腰1-6(L1-6)和右侧股骨的骨矿含量(BMC)和骨密度(BMD);放射免疫法(RIA)和酶联免疫法(ELISA)分别检测血清雌二醇(E2)、胰岛素样生长因子(IGF)及其结合蛋白(IGF-BP)含量。结果模型组雌鼠L1-6和股骨的BMC、BMD明显减少,血清E2、IGF-Ⅰ和IGF-BP1含量下降;雌激素和更年安怡片可不同程度地增加E2、IGF-Ⅰ含量(P<0.05或P<0.01),但BMC在用药前后无明显变化。更年安怡片预防组对上述指标无明显改善。结论更年安怡片可以通过上调E2、IGF-Ⅰ的含量,增加腰椎和股骨BMD,改善因去除双侧卵巢引起的实验性绝经后骨质疏松症,为临床运用更年安怡片治疗绝经后骨质疏松提供了实验依据。Objective To study the role of GNAT on treatment and prevention in postmenopausal osteoporosis female rats. Methods 60 female SD rats were divided randomly into six groups: intact ( INT), sham operation (Sham), model, estrodiol (E2), GNAT and GNAT prevention (GNAT PR) group. The bone mineral content (BMC) and the bone mineral density (BMD) of lumbar 1-6 and right thigh-bone were examined, as well as estrodiol (E2 ), insulin-like growth factor Ⅰ (IGF-Ⅰ) and insulin-like growth factor binding proteins (IGF-BPs) were tested by RIA or ELISA. Results The BMC and BMD of lumbar 1-6 and right thigh-bone in model group decreased obviously, meanwhile the serum E2 , IGF-Ⅰ and IGF-BP1 reduced too. E2 or GNAT could increase the BMD as well as upgrade the contents of E2 and IGF-Ⅰ . However, changes were not showed in above indexes in GNAT PR group. Conclusions GNAT could improve postmenopausal osteoporosis of female rats by upgrading the serum E2 and IGF-Ⅰ to increase BMD of postmenopausal osteoporosis rats, which provided an experimental evidence of GNAT in clinic.
