摘  要:IQ基序是与真核细胞内Ca^2+调控有关的重要结构域,具有特征性的核心序列和空间构象。它可特异性地募集和结合靶蛋白,后者从属于EF-hand蛋白家族,其中最重要的是钙调素,借此可调控胞内Ca^2+浓度变化,从而参与调节细胞Ca^2+信号转导途径。IQ基序与靶蛋白的结合受诸多因素和环节的影响,尤其是Ca^2+自身浓度变化对其可有反馈效应。目前已发现并报道的含IQ基序的蛋白广泛存在于各类生物体,定位于各级亚细胞结构,调控诸多生理和代谢过程,如肌纤维的收缩与舒张、离子通道状态、酶活性、细胞周期的维持、细胞骨架的组建以及生殖和发育等。对IQ基序结构特征和生物学功能进行简要阐述,有助于深入研究具有重要功能的含IQ基序蛋白和探寻新的未知蛋白。IQ motif, as a key domain to regulate Ca^2+ , was characterized by a core sequence and tridimensional structure. It could selectively recruit and bind target proteins, which belonged to EF-hand protein families. Among them, the one of great importance was calmodulin. The interaction between IQ motif and ealmodulin could definitely modulate the intraeellular concentration of Ca^2+ , which led to the regulation on the Ca^2+ signaling pathway. The binding affinity was also affected by a number of factors and processes, especially the concentration of Ca^2+ itself could act in a feedback loop. To date, all the reported IQ motif containing proteins were found in a large variety of species and subcellular organs, indicating a vast realm of physiological and metabolic regulation, for instance, the muscle contraction and relaxation, status of ion channels, enzymes' activity, maintenance of cell cycles, organization of cell skeleton, reproduction and development etc. Generally summarizing the structural characteristic and biological functions of IQ motif, would help further research on important IQ motif containing proteins and even exploration of new ones.
