摘 要:目的:从细胞凋亡角度探讨早期自然流产发病机制。方法:标本取自在空军总医院妇科就诊行流产的患者,自然流产组蜕膜及绒毛标本23例,并以正常早孕蜕膜及绒毛标本22例为对照组。采用TUNEL法检测细胞凋亡,免疫组化SP法检测p53、Bax及Bcl-2蛋白的表达。结果:与对照组比较,自然流产组蜕膜及绒毛组织细胞凋亡指数均明显增加(P<0.01),p53及Bax蛋白的表达也均明显增加(P<0.01),而Bcl-2蛋白的表达明显降低(P<0.01)。结论:细胞凋亡过度为自然流产发病机制之一,p53通过Bax/Bcl-2途径参与了自然流产蜕膜及绒毛组织细胞的凋亡调节。Objective To investigate the relationship between apoptosis and pathogenesis of spontaneous abortion at an early stage. Methods The specimens of decidua and chorionic villi were collected from 23 cases of spontaneous abortion at an early stage (abortion group) and 22 normal early pregnancies (control group) in our hospital. Apoptosis was detected by terminal deoxynucleotidy transferase-mediated dUTP-biotin nick end labeling (TUNEL), so were the expressions of p53, Bax, and Bel-2 proteins by immunohistochemistry (SP method). Results As compared with those in the control group, the apoptotie index and the expressions of p53 and Bax proteins in the deeidua and ehorionie villi tissues increased significantly in the abortion group (P 〈 0.01 ), while the Bel-2 protein expression decreased markedly (P 〈 0.01 ). Conclusions Excessive apoptosis is one of the mechanisms in the pathogenesis of spontaneous abortion. p53 is involved in the regulation of apoptosis of spontaneous abortion by the Bax/Bel-2 pathway.
