摘 要:[目的]为改造磷酸烯醇式丙酮酸羧化酶(PEPC)基因、提高PEPC活性寻找线索。[方法]对GenBank上登录的PEPC的cDNA序列、氨基酸序列进行BLAST搜索,运用生物信息学方法进行信号肽分析、亚细胞定位、结构域分析以及PEPC蛋白质氨基酸序列的同源性分析和系统进化树分析。[结果]共搜寻到62个PEPC蛋白质序列,绝大多数属于C3植物,9个属于C4植物。绝大多数PEPC没有存在信号肽的可能性。62个PEPC蛋白的亚细胞定位基本一致,均具有Phosphoenolpyruvate carboxylase结构域,未发现其他结构域。通过进化树分析可分别将C3植物和C4植物的PEPC酶分成4组和3组。62个PEPC蛋白质氨基酸序列的同源性为86.70%,而其在19个C3植物和9个C4植物中则分别为89.63%和89.61%。[结论]C4植物PEPC的423位上的丙氨酸和862位上的酪氨酸高度保守,而C3植物的428位和871位的氨基酸则缺少同源性,可能与C3和C4植物PEPC的活性差异有关。[Objective] The research aimed to search clues for modifying phosphoenolpyruvate earboxylase (PEPC) gene and increasing the activity of PEPC. [ Method] BLAST search was conducted on eDNA sequence and amino acid sequence of PEPC registered on GenBank website. Signal peptide analysis, subcellular localization, domain analysis, the homology analysis and phylogenetic tree analysis on amino acid sequences of PEPC protein were conducted by using the methods of bioinformatics. [ Result ] 62 sequences of PEPC protein were searched, most of them belonged to C3 plant and 9 of them belonged to C4 plant. Most of PEPC has no possibility of existing signal peptide. The subeellular localizations of 62 PEPC proteins were basically accordant and all had the domain of Phosphoenolpyruvate carboxylase and other domains were not found. PEPC enzymes in C3 plant and C4 plant could be respectively divided into 4 groups and 3 groups by the phylogenetic tree analysis. The homology of amino acid sequences in 62 PEPC proteins was 86.70% and that in 19 C3 plants and 9 C4 plants were 89.63 % and 89.61% resp. [ Conclusion] Alanine on 423 site and tyrosine on 862 site in PEPC of C4 plant were highly conservative, but the amino acids on 428 site and 871 site in PEPC of C3 plant were lacking of homology, which might be related with the differences of PEPC activity between C3 plant and C4 plant.
