摘  要:用大肠杆菌脂多糖(LPS)所致小鼠胸腺细胞凋亡模型和琼脂糖凝胶电泳、流式细胞术及免疫组织化学等方法,研究不同浓度高丽人参水提取物体内对小鼠胸腺细胞的影响。结果表明,每只小鼠腹腔注射10~20mg人参水提取物可防止LPS所致小鼠体重及胸腺减重,提高胸腺细胞存活力,抑制胸腺细胞DNA降解和促凋亡蛋白Bax的过量表达,免疫组织化学研究结果在蛋白水平上初步解释了人参抑制小鼠胸腺细胞凋亡的作用机制。Defective regulation of apoptosis may play a important role in the aetlology of cancer, AIDS, autoimmune diseases, and degenerative diseases of the central nervous system. The pharmacological manipulaiton of apoptosis offers new possibilities for the prevention and treatment of these lllnesses. ginseng, one of the valuable traditional chinese medicine, has many pharmacologic effects. In present study, we injected (i.p.) ginseng extract (GS) to the BALB/c mice administrated with bacterial lipopolysaccharide (LPS) and observed the effects of genseng on thymus weight, cell viability and DNA fragmentation confirmed by agarose gel electrophoreses, flow cytometry (FCM) and expressioin of Bax protein by immunocytochemistry. The results shown that (1) LPS could cause the decreasion of thymus weight, thymocyte number and viability and induced thymocytic apoptosis in vivo. (2) There were no significant difference between ginseng control group and normal control group. (3) Ginseng could increase the thymus weight, thymocyte number and viability of the mice administrated with LPS.(4) DNA fragmentation and apoptotic thymocytes iin the thymus of GS+LPS group was less than that in LPS group. (5) By immunocytology and image analysis, staining of bax protein in thymocytes of GS+LPS group was weaker than that in LPS group. These results indicate that ginseng could inhibite thymocytic apoptosis of the mice administrated with LPS.
