摘 要:目的观察中药菟丝子水提取物诱导毛囊无色素黑素细胞(amelanotic melanocytes,AMMC)分化的作用。方法以8-甲氧补骨脂素(8-MOP)作为阳性对照,选择系列浓度的中药菟丝子水提取物和8-MOP作用于体外培养的AMMC,然后测定酪氨酸酶活性、黑素含量和细胞增殖率的变化。透射电镜观察药物作用前后AMMC超微结构的变化。结果菟丝子水提取物明显促进AMMC生成黑素(P<0.01),且呈浓度依赖性,但是促进作用弱于阳性对照8-MOP(P<0.01)。电镜结果显示,无药物作用的AMMC主要含有I,II和III期黑素小体,药物作用以后,AMMC内出现了大量的IV期黑素小体。菟丝子水提取物对细胞增殖无明显影响,但明显增强酪氨酸酶的活性。结论菟丝子水提取物促进了AM-MC的分化,这种作用与其增强酪氨酸酶活性有关。Objective To investigate the effects of aqueous extracts of Cuscuta chinensis Lain on the differention of amelanotic melanocytes (AMMC) of outer root sheath from human hair follicles. Methods AMMC were incubated with different concentrations of aqueous extracts of Cuscuta chinensis Lain and 8-MOP, then activity of tyrosinase, melanin content and rate of proliferation of AMMC were tested respectively. Transmission electron microscopy was employed to observe the change of ultrastructure of AMMC after treatment. Results The aqueous extracts of Cuscuta chinensis Lain promoted melanogenesis of AMMC in close-dependent fashion (P 〈 0.01 ) ,but the effect was weaker than that of 8-MOP (P 〈 0.01 ). The melanosomes predominantly in stages Ⅰ,Ⅱor Ⅲwere observed in the cytoplasm of AMMC by TEM. After treatment with aqueous extracts of Cuscuta chinensis Lam the cells showed melanosomes mainly in stages Ⅲ and Ⅳ. In addition, the aqueous extracts of Cuscuta chinensis Lam enthanced tyrosinase activity of AMMC ,but showed no effect on proliferation of the cells. Conclusion The aqueous extracts of Cuscuta chinensis Lam could induce the differentiation of AMMC. The mechanism is related to activation of tytosinase of AMMC.
