摘  要:目的观察移植小胶质细胞是否可聚集在大鼠脑内β淀粉样蛋白(Aβ)周围并吞噬Aβ颗粒。方法在大鼠左右侧海马内分别注射Aβ42与生理盐水,3d后经颈内动脉将体外培养的持续表达增强型绿色荧光蛋白(enhanced green fluorescent protein,EGFP)的小胶质细胞注入大鼠体内,小胶质细胞注射后3d分别计数双侧海马内荧光细胞并在共聚焦显微镜下观察移植小胶质细胞是否吞噬Aβ颗粒。结果移植小胶质细胞聚集在Aβ沉积物周围,荧光显微镜下计数Aβ注射侧的EGFP阳性细胞数明显高于生理盐水注射侧的细胞数(P<0.05)。部分移植小胶质细胞吞噬Aβ沉积物,细胞内出现Aβ信号。结论移植小胶质细胞聚集在大鼠海马区Aβ沉积物周围,并摄取部分Aβ。Objective To evaluate whether exogenous microglia can gather around amyloid beta-protein (Aβ) deposit and phagocytize Aβ particles in the brain of rat model of Alzheimer's disease (AD). Methods Aβ42 and saline were injected into the left and right hippocampi of the rat respectively. Three days later, exogenous microglia consistently expressing enhanced green fluorescent protein (EGFP) were injected into the body of the rat by internal carotid artery. Three days after injection of microglia,the number of fluorescent cells in bilateral hippocampi of the rat were counted to observe whether exogenous microglia phagocyfize Aβ particles under confocal microscopy. Results Intra-arterially injected microglia positive for EGFP had accumulated at sites of Aβ deposition in the rat hippocampus. Few of exogenous microglia were seen in the region of saline injection. Some of the transplanted microglia phagocytized Aβ deposit with intraceUular Aβ signal. Conclusions Intra-arlerially injected exogenous microglia could accumulate around Aβ deposits in the hippecampus of rat and take in some distributed Aβ.
