PCR-RFLP Analysis of Three Wild Bananas and Their Resistance to Fusar-ium Wilt

摘  要:利用基于rDNA内转录间隔区(ITS)的PCR-RFLP标记和RAPD技术对采自深圳的3种野生蕉遗传背景进行了分析,并采用与香蕉枯萎病连锁的RAPD标记及苗期接种鉴定对其枯萎病的抗性进行了研究。研究结果表明野生蕉1号和3号为AA类群;野生蕉2号为BB类群。在遗传相似性系数为0.32处,可将同属AA的野生蕉1号和3号与贡蕉(AA)和巴西蕉(AAA)聚为一类;野生蕉2号(BB)与粉蕉(ABB)和大蕉(ABB)聚为一类。与香蕉枯萎病连锁的RAPD标记分析以及香蕉枯萎病的早期诊断结果均表明野生蕉2号和3号感香蕉枯萎病,野生蕉1号抗枯萎病。We used PCR -RFLP based on restriction-site variations in the internal transcribed spacers (ITS) of the nuclear ribosomal DNA genes and RAPD markers to assess the genetic diversity background of three wild bananas from Shenzhen. Moreover, three RAPD primers reported previously to be related to banana fusarium wilt and early screening techniques for Fusarium wilt resistance were used to analyses Fusarium wilt ( race 4) resistance of these wild bananas. The result of PCR - RFLP marker analysis indicated that genotype of wild banana 1 and wild banana 3 belonged to AA, but wild banana 2 was BB. The cuhivars for RAPD marker analysis included‘ Fenjiao' ( ABB), ‘ Gongjiao' (AA) , ‘ Baxijiao' ( AAA),‘ Longyajiao' (AAB) and ‘Dongguan Dajiao' (ABB) , when genetic similarity coefficient was set at 0. 32, wild banana 1 (AA) , 3 ( AA), ‘ Gongjiao' and ‘ Baxijiao' could fall into the same group ; the relationship between wild banana 2 ( BB), ‘ Fenjiao' ( ABB), and ‘ Dongguan Dajiao' (ABB) could classify into the same groups. Results of RAPD markers showing banding patterns specific to susceptible or resistant bananas and early screening technique for Fusarium wilt ( race 4) resistance both demonstrated that wild banana 2 and 3 were susceptible and wild banana 1 was resistant.
