摘　　要：目的 采用高效液相色谱法测定大鼠血浆中醋氯芬酸浓度，研究醋氯芬酸结肠靶向微丸在大鼠体内的药物动力学。方法 分别按低、中、高剂量（5、10、20mg·kg^-1）给予SD大鼠醋氯芬酸结肠靶向微丸并定时采血测定血药浓度。应用3P97药动学软件计算药动学参数。结果大鼠灌服低、中、高剂量醋氯芬酸结肠靶向微丸后主要药动学参数Cmax分别为（1．37±0．50）、（3．26±0．98）和（7．11±2．13）μg·mL^-1 tmax分别为（2．8±0．7）、（3，2±0．6）和（3．1±0．9）h，AUC0～24分别为（16．24土7．35）、（34．18±12．55）和（70．46±26．23）μg·mL^-1·h。结论高效液相色谱法准确、灵敏、简便，适用于醋氯芬酸血浆浓度的测定和药动学研究，醋氯芬酸结肠靶向微丸在大鼠体内呈线性动力学规律。Objective To determine the pharmacokinetics of aceclofenac colon specific pellets in rats. Methods Three different doses of aceclofenac colon specific pellets were given to 3 groups of rats. The drug concentration in the plasma was determined by HPLC and the data were processed with 3P97 software. Results The C t and AUC0～24 of aceclofenac colon specific pellets in rats with different doses were （1.37±0.50）, （3.26±0.98） and （7.11±2.13） /μg· mL^- 1 （2.8±0.7）, （3.2±0.6） and （3.1±0.9） h, （16.24±7.35）, （34.18±12.55） and （70.46±26.23） /μg · mL^-1· h, respectively. Conclusion The method is accurate, sensitive, simple and reproducible for the measure- ment of plasma concentration and pharmacokinetics in rats.
