摘 要:目的观察低氧高二氧化碳环境下,小鼠右心室收缩压(RVSP)、右心肥大指数的改变及肺小动脉结构的重建,初步建立小鼠低氧高二氧化碳性肺动脉高压模型。方法将C57BL/6小鼠随机分为实验2周组、实验4周组和对照组。实验组饲养于氧舱中,舱内氧浓度为9%~11%,二氧化碳浓度为5%~6%,每天8h,每周6d。实验2周组在舱内2周,实验4周组在舱内4周。至相应时间后检测RVSP、右心室与左心室+室间隔重量比[RV/(LV+S)]、肺小动脉管壁厚度与血管外径比(MT%)、血管壁面积与血管总面积比(MA%),并观察右心室心肌组织的病理改变。结果实验2周组小鼠RVSP、RV/(LV+S)、MT%、MA%均高于对照组(P〈0.05),并且光镜下可见心肌增生肥大。实验4周组小鼠的上述病变更为明显。结论在常压低氧高二氧化碳环境中2周即可形成较理想的肺动脉高压小鼠模型,这为更好地研究肺心病提供了新的途径。Objective To investigate the alterations in right ventricular systolic pressure (RVSP) and pulmonary vascular remodeling in mice exposed to isobaric atmosphere with low concentration of O2 with high concentration of CO2. Methods C57BL/6 mice were randomly divided into 2w exposure group.4w exposure group and control group. When they were exposed to an atmosphere containing 9%-11% O2 with 5%-6% CO2 for corresponding time respectively, RVSP, the right ventricular (RV) weight, the weight of left ventricle plus ventricular septum and pathological changes of the hearts and lungs in the mice were observed. Results RVSP and RV mass increased significantly after exposure for 2w and 4w compared with controls (P〈0.05). Pathology in the hearts and lungs of 2w group and 4w group also changed significantly compared with that of control group. Conclusion The mouse model of pulmonaly hypertension has been established, which can be used in COPD study.
