摘  要:报道利用高温溶剂法在Sinopak-S大孔硅胶上键合十八烷基,得到可用于蛋白质分离的高效反相色谱填料,合成时间缩短为3~5h。用苯、萘、菲及苯胺评价了该反相柱,其理论塔板数达每米3到5万,能有效地用于蛋白质和多肽混合物的分离。An improved method was presented for the synthesis of C18 reversed-phase chro- matographic column packing material based on Sinopak-S, a macroporous silica gel. The packing material was synthesized in 3 to 5 h under a higher temperature in Suitable solvents. The material showed a high carbon content on the silica gel Surface and excellent chro- matographic performance. Symmetrical peaks were obtained for the separation of benzene, naphthalene, phenanthrene, aniline and pyridine with a mobile phase of 85:15 methanol/ water solution. 3-5×104 theoretical plates/meter for the reversed-phase column was ob- tained. The column also showed an excellent ability for the separation of proteins and pep- tides in a 30 min-Iinear gradient program from 10 to 80% acetonitrile phosphate buffer solu- tion(pH3.2).
