长春花培养细胞中阿玛碱提取及高效液相色谱定量测定

摘　　要：建立固相快速提取及高效液相色谱测定长春花细胞培养物中阿玛碱的方法 ,并用此法对三种长春花细胞系 C2 0 hi,C2 0 D和 C2 0 的生物碱合成能力做了比较。研究发现 ,提取液 p H值在3.55～ 8.0范围内阿玛碱的回收率均在 95%～ 10 5%。荧光检测器的灵敏度可以达到 0 .4 5ng,进样量在 5～ 75ng之间时与峰面积呈线性关系 ,三种长春花细胞系的生物碱合成能力大小依次是 C2 0 hi>C2 0 D>This paper discribed a procedure for rapid extraction and quantitative assay of ajmalicine from cell suspensions of Catharanthus roseus and compared the capacity of alkaloid production of three cell lines C 20 hi,C 20 D and C 20 .The results indicate that ajmalicine recovery is between 95% and 105% when the extract pH value is from 3.55 to 8.0.With fluorescence, the detection limit of 0.45ng and a linear correlation between injected quantities and peak area were observed when the inject weight is between 5ng and 75ng. The sequence of alkaloid production of Catharanthus roseus cell lines was C 20 hi>C 20 D>C 20 .
